Now the fastest speedrun is Angerfist's 06:21.08 however it's tool-assisted, he takes damage and he misses all the secrets.
Also, he could have killed the bosses a bit faster.
Here's the goals for for this challenge.
No Damage = Perfect Bonus
Find all Secrets = Secret Bonus
Return to level start before entering the Exit Door = More enemies, more points!
Here's a few tips that might give you a bigger challenge, and not just focusing on speed.
1. More Enemies
When you've reached the end of a level, try going back to the start again. (Is there a term for this?)
You might encounter a few more enemies, such as in the first level, 3 green ninjas instead of 1.
In Round 4, level 1 if you take the water route, an additional enemy with a Bo will appear in the end.
In Round 1, level 2 it's possible, but tricky, to climb back up once you've fallen down to the fire poles.
You just gotta time it right.
It'll give you a few more Red Ninjas.
In general, backtracking brings more fun and exploration to the game. And more points too!
2. Flying Cannonball Bug
You can gather additional speed if you run down slopes such as Round 4, level 1.
If you run down the slope when the cannonball is on the left edge on the screen you can get it to travel along with you.
I dunno what you can use it for but maybe you can find some more glitches through that bug.
3. Water Sword Glitch
If you use the sword just before jumping into the water, you might fall to the bottom of the screen, ignoring the floating animation.
When you hit the bottom you'll start swimming again.
I haven't confirmed what causes this, maybe it's just at a specific place or maybe you need the power sword power-up first.
You might have to tap the attack button twice and hit the water surface just in-between attacks.
4. Ladder Warp
The biggest and worst cheat of all.
However, if you use it at the top of Round 1, level 2, you'll warp even higher than the intended game area, entering random block heaven.
Try it out! You might find something cool.
That's all the glitches I know of so far.
Try using these glitches in other parts of the game than I've mentioned and let me know if you find something cool!

Running backwards and forwards on the first level makes it possible to create a train of Red Ninjas. See how many you can gather!
